Asn336 is involved in the substrate affinity of glycine oxidase from Bacillus cereus Electron. J. Biotechnol.
Wu,Gaobing; Zhan,Tao; Guo,Yiming; Kumar,Ashok; Liu,Ziduo.
Background: Glycine oxidase (GO), a type of D-amino acid oxidase, is of biotechnological interest for its potential in several fields. In our previous study, we have characterized a new glycine oxidase (BceGO) from Bacillus cereus HYC-7. Here, a variant of N336K with increased the affinity against all the tested substrate was obtained by screening a random mutant library of BceGO. It is observed that the residue N336 is invariable between its homogeneous enzymes. This work was aimed to explore the role of the residue N336 in glycine oxidase by site-directed mutagenesis, kinetic assay, structure modeling and substrate docking. Results: The results showed that the affinity of N336H, N336K and N336R increased gradually toward all the substrates, with increase...

Bernardes,Patrícia Campos; Andrade,Nélio José de; Ferreira,Sukarno Olavo; Sá,João Paulo Natalino de; Araújo,Emiliane Andrade; Delatorre,Deyse Maria Zanom; Luiz,Lívia Maria Pinheiro.
The interaction between the surface of stainless steel and Bacillus cereus was studied in terms of the characteristics of interfacial interaction determined from the measurement of the contact angle of the surface of B. cereus and stainless steel in the presence or absence of B. cereus adherence. The microtopographies and the roughness of the surface of stainless steel and stainless steel adhered by B. cereus were evaluated with the help of atomic force microscopy and perfilometry. The strain of B. cereus studied was considered hydrophilic, whereas the stainless steel was considered hydrophobic. The adhesion was not thermodynamically favorable (ΔGadhesion > 0) between the stainless steel and the strain of B. cereus studied. Thus, the interaction between...

ABSTRACT Disease in agricultural field is a big problem that causes a massive loss in production. In this present investigation, we have reported a soil-borne bacterium Bacillus cereus IB311 which is antagonistic to plant pathogens (Pseudomonas syringae and Agrobacterium tumefaciens), and could make a substantial contribution to the prevention of plant diseases. To prove the practical application, the strain was directly applied in agricultural field. The results demonstrated that B. cereus IB311 has increased the production (20% and 26% in term of average pod number per plant, average seed number per pod, and seed yield per experimental plot) in ground nut (Arachis hypogaea var. Koushal, G201) and sesame (Sesamum indicum var. Kanak), respectively. To...

Biological control of Mycosphaerella fragariae in strawberry culture Sci. Agrar.Paran. / SAP
Heling, Anderson Luis; Kuhn, Odair José; Stangarlin, José Renato.
The Mycosphaerella spot is one of the main foliar diseases of strawberry, degrating great leaf regions and reducing the photosynthetic area. Its control is mainly by the use of chemical fungicides, but, due the increasing demand for food free of pesticide, alternative control methods have been researched, such as biological control. This work aimed to evaluate the effect on strawberry plants, treated with the biological control agents Bacillus cereus, Saccharomyces boulardii and Saccharomyces cerevisiae, in the severity of Mycosphaerella fragariae, productivity and in the activity of β-1.3 glucanases, peroxidases and chitinases enzymes. It was verified that S. cerevisiae and B. cereus treatments were similar to fungicide for disease control. However, even...

Enhancement of biohydrogen production from starch processing wastewater and further inside its ecosystem disclosed by 16S rDNA sequencing and FISH BABT
Sutthipattanasomboon,Chonlapin; Wongthanate,Jaruwan.
ABSTRACT Biohydrogen production from starch processing wastewater in this study resulted the highest yield of 61.75 mL H2/g COD at initial pH 7.0, thermophilic temperature, and iron concentration 800 mg Fe/L. The yield was 2-folded higher than the operation at mesophilic temperature or without iron addition. Cell immobilization by addition of biomaterials (BM) could improve the hydrogen yield by 2-folded comparing to the non-addition. BM from plants (loofa sponge) was found producing higher yield than that from animals (silk cocoon), and optimal concentration of BM was 5% (V/V). Furthermore, it was revealed further inside its ecosystem using SEM, 16S rDNA sequencing and FISH. There was found rod-shaped microorganisms of Bacillus cereus, which reported as...

Growth of enterotoxin producing Bacillus cereus in meat substrate at 10ºC and 30ºC BJM
Soares,Celina Mara; Kabuki,Dirce Yorika; Kuaye,Arnaldo Yoshiteru.
The behaviour of enterotoxin-producing Bacillus cereus in meat was investigated by inoculating spore suspensions of five cultures into meat substrate (pH 5.8) and incubating at 10ºC and 30ºC. The bacterial populations were evaluated after different times by plate counts in nutrient agar. All the cultures presented growth at 30ºC with the generation time varying from 28.8 to 36.0 minutes. Three cultures also presented growth at 10ºC with generation times between 10.16 and 28.38 h. Considering the results, it was concluded that meat kept at abusive temperatures would be subject to development of this microorganism.

Identification and characterization of the endophytic plant growth prompter Bacillus Cereus strain mq23 isolated from Sophora Alopecuroides root nodules BJM
Zhao,Longfei; Xu,Yajun; Sun,Ran; Deng,Zhenshan; Yang,Wenquan; Wei,Gehong.
Endophytes MQ23 and MQ23R isolated from Sophora alopecuroides root nodules were characterized by observing their ability to promote plant growth and employing molecular analysis techniques. Results showed that MQ23 and MQ23R are potential N2-fixing endophytes and belong to the same species as Bacillus cereus. MQ23 was shown to be able to produce siderophores, IAA, and demonstrate certain antifungal activity to plant pathogenic fungi. Co-inoculation with MQ23+MQ23II showed a more significant effect than inoculation alone in vitro for most of positive actions suggesting they have a cooperative interaction. Results of plant inoculation with endophytes indicated that the growth indexes of co-inoculated MQ23+MQ23II were higher than those of inoculated alone...

Interference in quality of strawberry caused by the use of biological agents for disease control Sci. Agrar.Paran. / SAP
Heling, A. L.; Favorito, P. A.; Kuhn, O. J..
The strawberry is a fruit of great market acceptance and it is much appreciated throughout the world, besides being very important for small farmers, generating employment and income. Nevertheless, this crop is affected by various diseases that require control, so that producers eventually use chemical fungicides, which, besides contaminating soil and water, eventually contaminate the fruit. In an attempt to reduce the amount of pesticides used in the strawberry crop, a lot of research on biological control of diseases has been done. However, the biological control of diseases in strawberries should not interfere negatively in fruit quality. Thus, this study aimed to evaluate the acidity, pH and total soluble solids (ºBrix) of strawberries treated with...
